Tip of the Month: Generate Sample Reports from Xcalibur Quan Browser

The Quan Browser is used to review results from quantitative analysis when using Thermo Scientific Xcalibur software. In Quan Browser you can review results, chromatograms of the target or quan ion, spectrum and calibration curves. When reviewing results in Quan Browser, you can make changes in both the integration and the calibration settings.

Proteomics: Proteome Discoverer 1.1 Software Released

Proteome Discoverer 1.1

For researchers working with proteins, revision 1.1 of the Thermo Scientific Proteome Discoverer software was released in December. It offers increased productivity through improved automation, batch processing, and support for large data sets. It also includes support for processing fractionated samples and MudPIT experiments, and for quantitation experiments using tandem mass tags (TMT). Clinical: Research Analysis of Clozapine and Norclozapine in Plasma using Automated Sample Preparation and LC-MS/MS

Clozapine is a tricyclic dibenzodiazepine drug used in the treatment of schizophrenia. It is uniquely effective in patients resistant to therapy with other antipsychotics. In addition to mandatory hematological monitoring to minimize the risk of agranulocytosis, there are large variations (50-fold) among patients’ clozapine dose requirements. Moreover, changes in smoking habits can have a large effect on the clozapine dose requirement (on average, the clozapine dose for non-smokers is half that required for smokers) due to the induction of cytochrome P450 (CYP) enzymes in smokers. Studies have indicated that accurate quantification of clozapine levels may help researchers better understand, and conduct analysis of, issues related to dose optimization and adherence.

This application note demonstrates the use of the automated TurboFlow method effectively eliminated two hours of sample preparation time for a 100-sample batch.

Environmental: Analysis of Sulfonamides in River Water using EQuan, an Online Concentration Analysis System

Triple stage quadrupole LC-MS systems are often used for highly sensitive quantitative analysis of environmental pollutants. The performance of an LC-MS system is one important factor that determines if it will efficiently and accurately detect pollutants present at low concentrations in environmental samples. Another factor is the process by which the system pre-treats the samples. In its sample pretreatment process, the Thermo Scientific EQuan system is able to quickly perform the time-consuming concentration process online, dramatically reducing the time required for analysis. Consequently, EQuan™ is capable of measuring multiple samples more efficiently than systems using off-line sample preparation, while reaching lower detection limits than are achievable with a conventional LC-MS/MS analysis.

Differential mobility spectrometry (DMS) systems, also referred to as high field asymmetric waveform ion mobility spectrometry (FAIMS) systems, enable identification of ions of interest before they are subjected to a mass spectrometer. Trap Talk: Improving the ETD Reagent Ion Intensity on Your Thermo Scientific Orbitrap

Many things can affect the reagent ion intensity on Electron Transfer Dissociation (ETD) instruments. If the reagent ion intensity drops below 5e5 in profile mode it is necessary to tune and/or calibrate the ETD components of the instrument. To improve the reagent ion intensity to a good level (generally > 1e6 in profile mode) follow this list of helpful hints.
